三、應(yīng)用
1、利巴韋林在紅細胞中磷酸化，生成三氮唑核苷單磷酸、二磷酸及三磷酸。其中三氮唑核苷單磷酸是肌苷單磷酸脫氫酶的強抑制劑，可抑制細胞鳥苷酸的合成，降低細胞鳥苷酸三磷酸化，阻斷病毒核酸的合成。
2、三氮唑核苷三磷酸抑制流感病毒RNA聚合酶，干擾病毒復制。
3、三氮唑核苷三磷酸抑制病毒mRNA5’末端的合成和轉(zhuǎn)錄酶，從而抑制DNA和RNA合成。對多種病毒 (包括DNA型病毒和RNA型病毒) 有抑制作用。對流感、腺病毒肺炎、甲型肝炎、皰疹、麻疹等有防治作用，對流行性出血熱也有效。
4、主要經(jīng)尿以原形排泄，也有少量脫糖三氮唑核苷代謝產(chǎn)物，少量從糞便排出。

1. Ribavirin is phosphorylated in red blood cells to produce ribavirin monophosphate, diphosphate and triphosphate. Among them, ribavirin monophosphate is a strong inhibitor of inosine monophosphate dehydrogenase, which can inhibit the synthesis of guanylic acid, reduce guanylic acid triphosphorylation, and block the synthesis of viral nucleic acid.
2. Ribavirin inhibits influenza virus RNA polymerase and interferes with virus replication.
3. Ribavirin inhibits the synthesis and transcriptase of the 5'-end of viral mRNA, thereby inhibiting DNA and RNA synthesis.
